Slice your STL into two pieces, add the first piece to the bed and add the second piece to it as a "part". Assign the first part to extruder 1 and the second part to extruder 2.

The M600 method PrusaSlicer uses doesn’t work with stock Ender firmware via Octoprint, it just gets ignored. Cura uses the M0 method that works well, but I haven’t found anything to replicate that in PrusaSlicer.
